Abstract: Platinhaltige Zytostatika, wie z. B. Cisplatin, werden in vivo durch eine Reihe von (bio-) chemischen Reaktionen verändert und metabolisiert. Die wichtigsten Prozesse dabei sind die intrazelluläre Hydrolyse und die Reaktion mit kleineren Nukleophilen oder Proteinen. Bezüglich des Verhaltens von Platinkomplexen im Cytosol sind einige Fragen weiterhin nur unbefriedigend geklärt, insbesondere hinsichtlich der Art und Bedeutung von Addukten des Platins mit intrazellulären Bindungspartnern. Im Rahmen dieser Arbeit wurde mit Hilfe des fluoreszierenden Cisplatinanalogons CFDA-Pt-Cl2 das Verhalten von Platinkomplexen im niedermolekularen Bereich des Cytosols einer Cisplatin-sensitiven und einer Cisplatin-resistenten Krebszelllinie untersucht. Die hierfür entwickelten und optimierten kapillarelektrophoretischen Trennmethoden erlauben es, niedermolekulare Platinspezies durch die Detektion der Fluoreszenz (CE-LIF) mit kurzen Analysenzeiten (unter fünf Minuten) empfindlich (pmol/L) und reproduzierbar nachzuweisen und zu quantifizieren und mittels Massenspektrometrie (CE-MS) zu identifizieren. Durch Kontroll- bzw. Modellversuche wurde die Reinheit des verwendeten CFDA-Pt-Cl2 sowie die Stabilität und Integrität der Zellkulturproben sichergestellt, die gesamte Probenvorbereitung optimiert und aufgezeigt, dass CFDA-Pt-Cl2 vergleichbare Komplexe, u.a. mit Glutathion, Cystein, Methionin, bildet wie Cisplatin. Die Behandlung von Zellen beider Zelllinien mit CFDA-Pt-Cl2 zeigte, dass der Dichlorokomplex die dominierende Platinspezies (über 70 %) in der niedermolekularen Fraktion des Cytosols ist. Eine Metabolisierung bzw. Hydrolyse des Dichlorokomplexes war nur teilweise und für beide Zelllinien in vergleichbarer Art und Weise zu beobachten. Unterschiede zwischen der Cisplatin-sensitiven und der Cisplatin-resistenten Krebszelllinie konnte bezüglich des Gehaltes an Platinspezies beobachtet werden. Bei Normierung auf den Proteingehalt konnte in den sensitiven Zellen ein höherer Gehalt an Platinspezies in der niedermolekularen Fraktion des Cytosols nachgewiesen werden als in den Zellen der resistenten Zelllinie.
Pt-based chemotherapeutic agents, such as cisplatin, are known to undergo several (bio-) chemical transformation steps after administration. Intracellular hydrolysis and ligand exchange or adduct formation with small nucleophiles and larger proteins are their most relevant reactions on the way to the final target (DNA), but there are still many open questions regarding the identity and pharmacological relevance of various proposed adducts and intermediates of cisplatin in the cytosol. In the present work the cellular interactions of platinum complexes were studied by using the fluorescent cisplatin analogue CFDA-Pt-Cl2. Cells of an ovarian cisplatin-sensitive cell line and its cisplatin-resistant variant were treated with CFDA-Pt-Cl2 and the low molecular fraction of the cytosol were analyzed by capillary zone electrophoresis. The developed and optimized methods enable a very sensitive (pmol/L) and accurate analysis of low molecular mass Pt species, involving a fast CE-LIF detection within 5 min and identification by CE-MS. Control and further model experiments ensure the purity of the CFDA-Pt-Cl2 as well as the stability and the integrity of the cell culture samples. The whole sample preparation was optimized and CFDA-Pt-Cl2 was checked for forming similar adducts as Cisplatin. After treating cells of both cell line with CFDA-Pt-Cl2 the di-chloro complex represented the predominant platin compound (70 %) in the low molecular fraction of the cytosol. Hydrolysis, ligand exchange or adduct formation was only observed for the minor part and occurred in both cell lines in a similar manner. Differences between both cell lines appeared regarding the platin levels. Normalized to the protein content higher levels of platin compounds were observed in the low molecular fraction of the cytosol in cells of the cisplatin-sensitive cell line.
