Abstract: Die vorliegende Dissertation beschäftigt sich mit der Entwicklung einer intramolekularen Alkin-(Aza-)de Mayo Reaktion, die zur Synthese von mittelgroßen anellierten cyclischen Dienonen genutzt werden kann. Als Ausgangsmaterial dienen einfach zugängliche Alkinyl-substituierte vinyloge (Thio-)Ester bzw. Imide. Entscheidend für die erfolgreiche Durchführung dieser photochemischen Reaktion ist die Verwendung einer quasi-monochromatischen Lichtquelle zur Bestrahlung. Des Weiteren begünstigt der Einsatz von polar-protischen Lösungsmitteln, vorzugsweise perfluoriert, den Ablauf der Reaktion. Es handelt sich bei der Transformation um eine photochemisch induzierte Zwei-Kohlenstoffatom-Ringerweiterung, mit der durch die Intramolekularität eine Anellierung der neu gebildeten Ringe einhergeht. Werden o-alkinylierten Anilinen zur Synthese der photoaktiven Substrate verwendet, kann durch die Anwendung der Alkin-Aza-de Mayo Reaktion das Strukturelement der Cyclohepta[b]indole aufgebaut werden. Das Cyclohepta[b]indol-Strukturmotiv ist in Naturstoffen und menschengemachten pharmakologisch relevanten Verbindungen oft vertreten. Des Weiteren können die erzeugten Dihydrocyclohepta[b]indolone verwendet werden, um die Stoffklasse der Indolo[2,3-d]tropone zugänglich zu machen. Die bislang unbekannten Indolo[2,3-d]tropone können strukturell den Heteroacenen zugeordnet werden.
The present thesis describes the development of an intramolecular alkyne-(aza-)de Mayo reaction for the synthesis of annulated medium-sized cyclic dienones, utilizing alkynylated vinylogous (thio-)esters or imides as the starting material. For the success of the photochemical reaction the application of a quasi-monochromatic light source is essential. Furthermore, the use of a polar protic solvent, preferably perfluorinated, has a beneficial effect on the reaction. The transformation can be seen as a photochemically triggered two-carbon ring expansion and, due to its intramolecularity, the process is accompanied with the annulation of two new formed rings. Using o-alkynylated anilines for the synthesis of photoactive substrates and deploying these compounds in the alkyne-aza-de Mayo reaction, a cyclohepta[b]indole motive can be established. The cyclohepta[b]indole structural motive often occurs in natural products and synthetic pharmacologically relevant compounds. With the dihydrocyclohepta[b]indolones in hand, it is possible to convert them into the corresponding indolo[2,3-d]tropones. The so far unknown indolo[2,3-d]tropones can be classified as heteroacenes.
