dc.description.abstractZur umfassenden Identifizierung und Quantifizierung aller in einer komplexen Probe befindlichen Analyten steht die analytische Chemie derzeit im Bereich der Massenspektrometrie vor großen Herausforderungen. Grund ist eine fehlende, geeignete Probenzufuhr-und Ionisierungsmethode. In den letzten Jahren hat sich hier die Elektrosprayionisierung durchgesetzt, da sie Probenzufuhr und Ionisierung in einem Aufbau vereint. Zudem arbeitet sie unter Umgebungsbedingungen und eignet sich deshalb besonders für meist temperaturempfindliche, biologische Proben, allerdings nur für die Analyse polarer Analyten. Mittel- bis unpolare Analyten können entweder gar nicht, oder nur durch eine aufwendige Probenvorbereitung analysiert werden. In dieser Dissertation wird eine neuartige, Plasma-basierte Ionisierungsquelle mit einem Elektrospray kombiniert, um eine universelle Ionisierung zu erzielen und die Analyse von komplexen Proben zu erleichtern. Dafür wurde das Flexible MicroTube Plasma (FµTP) als Plasma-basierte und miniaturisierte Ionisierungsquelle ausgehend von einer dielektrisch-behinderten Entladung, grundlegend neuentwickelt und in Bezug auf die elektrische Feldstärke, den Potentialgradienten, die lineare Gasgeschwindigkeit, die Anwendbarkeit für die Analytik und die reproduzierbare Fertigung optimiert. Im Anschluss ermöglichte das FµTP-Design eine erfolgreiche Kombination mit einer nano-Elektrosprayionisierung und bietet erstmals die Möglichkeit zur simultanen Analyse von Cholesterin und verschiedener Diacyl-Glycerine sowie Phosphatidyl-Choline innerhalb einer komplexen Leberprobe.de
dc.description.abstractCurrently, analytical chemistry is facing major challenges in the mass spectrometry field to identify and quantify all analytes of a complex sample comprehensively. Therefore, a suitable sample feed and ionisation system is crucial. In recent years, electrospray ionisation has asserted itself, because it enables the combination of the sample feed and the ionisation system. Moreover, electrospray ionisation works under ambient conditions and is thus particularly suitable for heat-instable biological samples, though only polar molecules can be analysed. Medium to non-polar analytes can either not be investigated at all or have to undergo a prior and extensive sample preparation. To circumvent this hindrance, this work presents a novel plasma-based ionisation combined with an electrospray ionisation to achieve a more comprehensive and universal ionisation, even for complex samples. Therefore, the design of a dielectric barrier discharge has been fundamentally redeveloped and optimised with regard to the electric field strength, the potential gradient, the linear gas velocity, usability, and standardised production resulting in a novel design of a plasma-based ionisation, namely the Flexible MicroTube Plasma (FµTP). Successfully, the FµTP enabled the direct combination with a nano-electrospray ionisation and for the first time, a simultaneous analysis of cholesterol, diacylglycerols, and phosphatidylcholines in a complex liver sample was possible.de
