Endgruppenfunktionalisierte Poly(2-R-2-oxazoline) zum Design neuartiger Hochpolymere, Netzwerke und antimikrobieller Polymere

dc.description.abstractPoly(2-R-2-oxazoline) (POx) bilden eine vielseitige Polymerisationsklasse und können für ein breites Spektrum an Anwendungsmöglichkeiten eingesetzt werden. Hierbei lag der Fokus bisher oft auf der Variation des Polymerrückgrats und es wurden diverse funktionelle Seitengruppen in die Polymerketten eingebaut. Da POx über eine lebende kationische Ringöffnungspolymerisation polymerisiert werden, können auch an den Polymerenden gezielt funktionelle Gruppen eingebaut werden. Dies eröffnet ein großes Potential für weitere Anwendungsgebiete von POx. Im Rahmen dieser Arbeit wurden POx mit diversen Endgruppen beidseitig ausgestattet, um neuartige Hochpolymere, Netzwerke und antimikrobielle Polymere zu designen. Zunächst wurden die POx mit Hydroxyl-, Amino-, Thiol- und Acrylatendgruppen versetzt, um Makromonomere mit zwei funktionellen Endgruppen zu gewinnen. Diese wurden dann in diversen Stufenreaktionen polymerisiert. So konnte gezeigt werden, dass sich POx mit lebenden Kettenenden direkt verlinken lassen und dass POx zu Polyurethanen, Polyestern und Polyamiden, sowie über Michael-Additionen polymerisiert werden können. Dabei konnten Hochpolymere mit Molekülargewichten von bis zu etwa Mn = 12000 g·mol-1 und bis zu 12 Makrowiederholungseinheiten erfolgreich hergestellt werden. Die bifunktionellen POx wurden ebenfalls als Makroverlinker mit Poly(2-ethylhexylacrylat) in amphiphilen Polymerconetzwerken (APCNs) eingesetzt. Durch die Untersuchung der in Wasser und Toluol gequollenen APCNs mittels Kleinwinkel-Röntgenstreuung (SAXS) ist es erstmals gelungen die Topologie der gequollenen Netzwerke zu bestimmen. Dabei hat sich gezeigt, dass nur die Poly(2-ethylhexylacrylat)-Phase ein homogenes Quellverhalten durch das gesamte Netzwerk hindurch aufweist. Die PMOx-Phase hingegen liegt teilweise isoliert im gequollenen Netzwerk vor. Diese Erkenntnis ist vor allem für die Anwendung von APCNs als Trägersysteme von Bedeutung. Werden Poly(2-methyl-2-oxazoline) (PMOx) mit quartären Ammoniumendgruppen ausgestattet, weisen sie eine hohe antimikrobielle Wirksamkeit gegen Gram-positive Bakterienstämme auf. Durch die Hydrolyse des Polymerrückgrats zu Polyethylenimin (PEI) konnte die antimikrobielle Wirksamkeit gegen Gram-negative Stämme erweitert werden. Die Untersuchung des Wirkmechanismus zeigte einen synergetischen Effekt zwischen dem Polymerrückgrat und den Endgruppen, welcher die antimikrobielle Wirksamkeit sowohl gegen Gram-negative als auch Gram-positive Bakterienstämme erhöht.de
dc.description.abstractPoly(2-R-2-oxazolines) (POx) are a versatile class of polymers with a broad spectrum of applications. So far the focus of studies on POx was laid on the variation of the polymer backbone and its numerus functional side chains. The polymerization mechanism of POx allows the introduction of functional end groups during the termination process. This enlarges the field of possible applications of POx. The aim of this work was to use POx with versatile functional end groups to design novel polymers with high molecular weights, networks, and antimicrobial polymers. Initially POx with hydroxyl, amino, thiol and acrylate end groups were synthesized to gain macromonomers with two functional end groups. These were polymerized in different step growth reactions. It could be shown, that POx with living end groups can be directly polymerized. Additionally POx could be used in polyurethane, polyester, and polyamide polymerizations, as well as in Michael additions to gain polymers with molecular weights of up to Mn = 12000 g·mol-1 and 12 macromer repeating units. Bifunctional POx was also used as macro cross linker with poly(2-ethylhexylacrylate) to form amphiphilic polymer conetworks (APCNs). By studying the networks in the swollen states in the orthogonal solvents water and toluene with small-angle X-ray scattering, it was possible to analyze the topology of the swollen networks for the first time. It could be shown, that only the poly(2-ethylhexylacrylate) phase swells homogenously throughout the whole network. Whereas the PMOx-Phase showed partially isolated phases in the swollen APCNs. Poly(2-methyl-2-oxazolines) (PMOx) with quaternary ammonium end groups are highly active against gram-positive bacteria. In this study, the polymer backbone was hydrolyzed to polyethyleneimine. This led to an additional activity against gram-negative bacterial strains. Analysis of the mode of action revealed a synergetic effect of the polymer backbone and the end groups, which increased the antimicrobial activity against both types of bacterial strains.en
