Abstract: Das Spiro[5.5]ketal ist die zu Grunde liegende Teilstruktur zahlreicher biologisch aktiver Naturstoffe. Aufgrund der Tatsache, dass vereinfachte, charakteristische Spiroketale, die von Naturstoffen abgeleitet worden waren, ihre biologische Aktivität behielten, zeigt, dass das Spiro[5.5]ketal als biologisch prävalidierte Struktur betrachtet werden kann und somit als Leitstruktur in der Naturstoff-basierten Bibliothekssynthese geeignet ist. In dieser Dissertation wird über zwei unterschiedliche Wege berichtet, Spiroketale stereoselektiv an der Festphase zu synthetisieren. Die erste Strategie geht von enantiomerenreinen Bausteinen aus, verwendet als wesentlichen Syntheseschritt eine doppelte, interne Hetero-Michael-Addition und führt zu den Spiro[5.5]ketalen in hoher Ausbeute und Reinheit. In einer zweiten Strategie wird die enantioselektive Synthese von Spiroketalen am polymeren Träger erreicht. Die Reaktionssequenz verwendet die asymmetrische, Bor-Enolat-vermittelte Aldolreaktion als stereodifferenzierenden Schritt, wobei das Bor-Enolat am Polymer gebunden ist oder in Lösung vorliegt. Sie verläuft über 12 Stufen an der Festphase und führt zu den gewünschten Spiroketalen in hoher Gesamtausbeute und hoher Stereoselektivität. Das Testen beider Bibliotheken ergab mehrere Verbindungen, die zum einen Phosphatasen inhibieren und zum anderen den Aufbau des Tubulingerüsts in Brustkrebszellen modulieren, ohne Mikrotubulin als direktes Ziel aufzuweisen.
The spiro[5.5]ketal forms the underlying structural skeleton of numerous biologically active natural products. Since simplified but characteristic spiroketals derived from the parent natural products retain biological activity, the spiro[5.5]ketal can be regarded as a biologically pre-validated framework for the development of natural product-derived compound collections. Within this thesis two routes were found to synthesize spiroketals in a stereoselective manner on the solid support. The first strategy starts from enantiomerically pure building blocks, applies a double internal hetero Michael addition as key step and gives the spiroketals in high overall yield and purity. In a second strategy the enantioselective synthesis of spiro[5.5]ketals on solid support is reported. The reaction sequence employs asymmetric boron-enolate aldol reactions with the enolate bound to the polymer or in solution as the key enantiodifferentiating step. It proceeds in up to 12 steps on solid support and makes the desired spiroketals available with high overall yield and with high stereoselectivity. The screening of the two libraries revealed both, compounds that inhibit phosphatases and compounds that modulate formation of the tubulin cytoskeleton in human cancer cells without directly targeting microtubules.
