dc.description.abstractDie Markierung von Proteinen mit physikalischen Sonden ist eine technische Herausforderung. Hierzu wurden eine Vielzahl teils aufwendiger Methoden entwickelt. Die derzeitige Standardmethode zur ESR Markierung von Proteinen ist das site directed spin labeling (SDSL). In dieser Arbeit wurde eine Alternative zum SDSL entwickelt. Als Methode zur Markierung wurde eine Kombination aus Festphasensynthese von Peptide und chemischer Ligation (NCL & EPL) verwendet. Der notwendig kompatible ESR Marker wurde entwickelt und in guten Ausbeuten synthetisiert. Der ESR Marker konnte in Peptide eingebracht und in den biologischen Systemen RBD/Ras und HtrII/SRII verwendet werden.de
dc.description.abstractLabeling of proteins with biophysical probes is a technical challenge. A large number of methods have been developed recently. The standard method for EPR labeling of proteins is site directed spin labeling (SDSL). An alternative for SDSL is shown here. The methods of choice for labeling of peptides are solid phase peptide synthesis and chemical ligation methods (NCL & EPL). A compatible EPR probe was synthesized in good yields. The EPR label was incorporated in peptides and the biological systems RBD/Ras and HtrII/SRII.de
