dc.description.abstractIm Rahmen dieser Dissertation wurde ein Baukastenkonzept zur Synthese photoaffinitätsmarkierter menschlicher N-Ras-Peptide und -Proteine entwickelt. Damit konnten maßgeschneiderte 'Sonden' für die Untersuchung der post-translationalen Modifikation der Ras-Proteine sowie ihrer Wechselwirkung mit anderen Proteinen bereitgestellt werden. Schlüsselschritt der Synthese ist die Kondensation Benzophenon-markierter, prenylierter Cysteinfragmente mit am polymeren Träger aufgebauten Peptiden. In biologischen Experimenten wurde die Gültigkeit des Konzepts unter Beweis gestellt.Darüber hinaus konnten Phenylhydrazide als enzymatisch mit Tyrosinase abspaltbare C-terminale Schutzgruppe in der Synthese von Peptiden und sogar Peptidkonjugaten etabliert werden. Die Abspaltung erfolgt bei neutralem pH und Raumtemperatur mit ausgezeichneter Chemo- und Regioselektivität und erwies sich als orthogonal zu den gängigen Carbamatschutzgruppen. Mechanistische Untersuchungen zeigten, dass bei der zweistufigen Abspaltung Phenylradikale freigesetzt werden.de
dc.description.abstractThis thesis describes the development of a modular construction kit approach for the synthesis of photoaffinity-labelled human N-Ras-peptides and -proteins. The method allows to synthesize 'tailor-made' probes for studying the post-translational modification of Ras-proteins and their interactions with other proteins. Key step of the synthesis is the fragment condensation of benzophenone-labelled prenylated cysteines with peptides synthesized on the polymeric support. Biological experiments furnished a proof of principle.Furthermore, phenylhydrazides were established as an enzyme-labile C-terminal protecting group in the synthesis of peptides and peptide conjugates. Their cleavage occurs under mild conditions at room temperature and neutral pH using tyrosinase isolated from mushrooms. The chemo- and regioselectivity of the biocatalyst is excellent, and the protecting group is orthogonal to the most important carbamate protecting groups. Mechanistic investigations revealed that the cleavage occurs in a two-step process liberating phenyl radicals.en
