dc.description.abstractIm Jahr 2001 berichteten KOBAYASHI et al. von der Isolierung des Daphnicyclidin Alkaloids Daphnicyclidin A und weiterer Vertreter dieser neuen Alkaloid Klasse aus den Scheinlorbeergewächsen Daphniphyllum Teijsmanni und Daphniphyllum Humile. Diese beeindruckenden Alkaloide zeichnen sich neben dem käfigartigen polycyclisch anellierten Ringsystem weiterhin durch ein einzigartiges Pentafulven-Strukturmotiv aus. Die vorliegende Arbeit beschreibt die 15-stufige Synthese eines ABC-tricyclischen Synthesebausteins ausgehend von Isoprenol. Als Schlüsselreaktionen zum Aufbau dieser Verbindung dienten eine Pd(0)-Enolat Alkenylierung zur Anellierung des B-Rings sowie die Kombination einer retro-MANNICH-Fragmentierung und einer unterbrochenen aza DE MAYO Reaktion eines tricyclischen Photoproduktes zur Konstruktion des AC-Ringsystems. Weiterhin konnte es gelingen, in einer ebenfalls 15-stufigen Synthese den Zugang zu einem ACD-tricyclischen Diketoimin zu etablieren. Schlüsselreaktion zum Aufbau dieser Verbindung war neben der Kombination aus retro-MANNICH-Fragmentierung und einer unterbrochenen aza-DE MAYO-Reaktion eines tetracyclischen Photoproduktes zur Konstruktion des AC-Ringsystems eine Zweikohlenstoff-Ringerweiterung eines β-Ketoesters. Mit Hilfe der unterbrochenen aza-DE MAYO-Reaktion konnte ein robuster und skalierbarer Zugang zu vier komplexen β-Aminocarbamaten etabliert werden.de
dc.description.abstractIn 2001, KOBAYASHI et al. reported the isolation of the daphnicyclidin alkaloid Daphnicyclidin A and other members of this new class of alkaloids from Daphniphyllum teijsmanni und Daphniphyllum humile. In addition to their cage-like polycyclic fused ring systems, these remarkable alkaloids are further characterized by a unique pentafulvene structural motif. This thesis describes the 15-step synthesis of an ABC-tricyclic synthetic building block starting from readily available isoprenole. The key reactions for the construction of this compound were a Pd(0)-catalyzed enolate alkenylation for the annelation of the B-ring, as well as the combination of a retro-MANNICH fragmentation and an interrupted aza-DE MAYO reaction of tricyclic photoproduct for the construction of the AC-ring system. Furthermore, we were able to establish a route to an ACD-tricyclic diketoimine in a 15 step synthesis. The key reaction for the construction of this compound was, in addition to the combination of retro-MANNICH fragmentation and an interrupted aza-DE MAYO reaction of tetracyclic photoproduct to construct the AC ring system, a two-carbon ring expansion of a β-ketoester. Using the interrupted aza-DE MAYO reaction, a robust and scalable route to four complex β aminoketones was established.en
