Title: Enantiomerenreine Tris(hydroxymethyl)ethan-Derivate als chirale Synthesebausteine und Synthese von (20R,22E)- und (20S,22E)-3-Oxochola-1,4,22-trien-24-säuremethylester

Abstract: Das Ziel des ersten Projektes war die Darstellung eines enantiomer einheitlichen Synthese-bausteins auf der Basis von Tris(hydroxymethyl)ethan. Über eine enzymkatalysierte Vereste-rung mit Chirazyme-L2 und Vinylacetat als Acyldonor wurde monotrityl-geschützes homo-chirales Tris(hydroxymethyl)ethan desymmetrisiert. Durch gezielte Optimierung der Reak-tionsbedingungen erhielt man die entsprechenden Monoacetate in Ausbeuten von 80 % mit Enantiomerenüberschüssen von bis zu 83 %. Diese wurden zur weiteren optischen Anreicher-ung in geeignete kristalline Derivate überführt. Durch zweimalige Umkristallisation von (R)-(-)-3-(4-Chlorbenzoyloxy)-2-methyl-1-(p-toluolsulfonyloxy)-2-(trityloxy-methyl)-propan war ein enantiomerenreiner Baustein dargestellt. Die Bestimmung der absoluten Konfiguration dieses Synthesebausteins gelang durch chemische Korrelation mit 1-Brom-3-chlor-2-hydro-xymethyl-2-methyl-propan. Durch die Darstellung beider enantiomerer Formen von 4-Formyl-4-methyl-tetrahydrofuran-2-on wurde die Einsatzfähigkeit des Bausteins in der organischen Synthese demonstriert.(22E)-3-Oxochola-1,4,22-trien-24-säuremethylester ist ein natürlich vorkommendes Steroid mit nicht spezifizierter Konfiguration an C-20. Im Rahmen eines zweiten Projekts wurde ein Gemisch der C-20-Epimere in 4 Stufen synthetisiert und anschließend durch Kristallisation getrennt. Beide Epimere wurden durch Röntgenstrukturanalyse charakterisiert und dem Naturstoff wurde die 20S-Konfiguration zugewiesen.
In a first project, a chiral building block, based on a tris(hydroxymethyl)ethane-derivative should be prepared. An enzyme-catalyzed transesterification, using Chirazyme-L2 and vinyl acetate led to the desymmetrized monoprotected homochiral tris(hydroxymethyl)ethane. Optimized reaction conditions gave the corresponding mono acetate in a yield of 80 % and with an enantiomeric excess up to 83 %. An enrichment of the optical purity was achieved by crystallization of a suitable cristalline derivative. The enantiomeric pure building block was obtained after two recristallizations of (R)-(-) 4-chloro-benzoic acid 2-methyl-2-(toluene-4-sulfonyloxymethyl)-3-trityloxy-propyl ester. Its absolut configuration has been determined by chemical correlation of 1-bromo-3-chloro-2-hydroxymethyl-2-methyl-pro-pane and its usability in Organic Synthesis has been demonstrated by the transformation in both enantiomeric forms of 3-methyl-5-oxo-tetrahydrofuran-3-carbaldehyde. Methyl (22E)-3-oxochola-1,4,22-trien-24-oate is a naturally occuring steroid with unknown configuration at C-20. In a second project, a mixture of the C-20 epimers has been synthesized in a four step procedure starting from (20S)-3-oxo-23,24-dinorchol-4-en-22-al. The diastereomeres were separated via fractional crystallization and caracterized by X-ray analysis. The configuration at C-20 of the naturally occuring steroid has been determined to be S.
