dc.description.abstractBedingt durch ein negatives Aktivierungsvolumen lassen sich Proteine durch Druckänderung reversibel entfalten. Mit Hilfe einer speziell für Drücke bis 6000 bar und Drucksprungampli-tuden bis 3200 bar ausgelegten Apparatur werden zeitabhängig Röntgenkleinwinkelstreudaten (TRSAXS) gemessen und die kinetischen Daten des Faltungsprozesses aus dem Gyrationsra-dius Rg bzw. der Paarabstandsverteilungsfunktion p(r) bestimmt. Weiterhin werden dreidi-mensionale Strukturvorschläge des gefalteten und entfalteten Zustands berechnet und mit Röntgenkristall- bzw. NMR-Strukturen verglichen. Die Verteilung von Sekundärstrukturele-menten aus FTIR-spektroskopische Daten werden herangezogen, um einen Mechanismus für die Strukturänderungen innerhalb der Proteine vorzuschlagen. Im Rahmen dieser Arbeit wer-den Alkoholdehydrogenase (LADH), Staphylokokken Nuklease (Snase) und der Tryptophan Repressor (Trp) untersucht. Der zweite Teil der Arbeit befasst sich mit dem Phasenverhalten und der Struktur von AOT-Mikroemulsionen. Hierzu werden Röntgen- und Neutronenkleinwinkelstreustudien unter Va-riation des Drucks p durchgeführt. Dabei wird die Bildung von neuen Strukturen, wie Stäbchenmizellen, lamellare und bikontinuierliche Phasen, beobachtet. Die Größenparameter lassen sich direkt durch die Annahme polydisperser sphärischer Partikel aus den Streudaten oder nach der Fourier-Transformation durch Vergleich der Korrelationsfunktion des Teubner-Strey-Modells mit der Paarabstandsverteilungsfunktion ermitteln. In die untersuchten Mi-kroemulsionen (invers-mizellare Phase) lassen sich Enzyme inkorporieren und Enzymreak-tionen durchführen. Die Reaktion von ?-Chymotrypsin mit einem Modellpeptid und LADH mit Alkohol zeigt, bedingt durch das negative Aktivierungsvolumen der Reaktion, eine Zu-nahme der Geschwindigkeit durch Erhöhung des Drucks. Die Kombination enzymkinetischer und struktureller Ergebnisse ergeben für ?-Chymotrypsin und insbesondere für Alkoholdehy-drogenase (LADH), welches ab einem Druck von 1500 bar durch Dissoziation inaktiv wird, einen eindeutigen stabilisierenden Einfluss durch den Einbau in inverse Mizellen. Im Gegen-satz zur Aktivität von LADH in wässrigem Milieu nimmt diese in mizellarer Umgebung ab 1500 bar drastisch zu, eine Inaktivierung durch Dissoziation wird somit unterbunden. Die Beschleunigung der Reaktion wird durch Bildung von Stäbchenmizellen begünstigt, die eine optimale Anpassung an die ellipsiodale Proteinstruktur bedeutet.de
dc.description.abstractCaused by the negative activation volume proteins can be unfolded reversibly through appli-cation of high pressure. A special apparatus was constructed and used to allow a pressure range up to 6000 bar and pressure-jumps with pressure amplitudes up to 3200 bar. The kinetic data of the folding process is monitored with Time-Resolved Small-Angle X-Ray Scattering (TRSAXS) and determined from the change of the radius of gyration Rg and the pair-distance distribution function p(r) with time. Additionally, three-dimensional model structures are cal-culated from the scattering data and compared with structural data determined by X-Ray Dif-fraction (XRD) and NMR. The distribution of secondary structure elements is taken from FT-IR data of the proteins and combined with structural information; a possible unfolding path-way is proposed. In this work the proteins alcohol dehydrogenase (LADH), staphylococcal nuclease (Snase) and tryptophane repressor (Trp) are studied. The second part of this work contains the examination of the phase behavior and structure of AOT microemulsions with Small-Angle X-Ray and Neutron Scattering. The application of pressure leads to transitions to new phases like rod-like micelles, bicontinuous and lamellar phases. The size parameters can be determined directly from the scattering data with model structure factors. A new way to obtain the size parameters of the observed phase is the com-parison of the Teubner-Strey correlation function with the pair-distance distribution function p(r). Enzymes can be incorporated in microemulsions and reactions with substrates are monitored to characterize the stability of enzymes and the influence of the surfactant mem-brane on the enzyme. The reaction of ?-chymotrypsin with a model peptide and alcohol de-hydrogenase (LADH) with alcohol shows an increasing activity with increasing pressure caused by a negative reaction volume. The combination of the kinetic data and the structural data reveal a definite proof of a stabilizing effect of inverse micelles on the examined en-zymes. Especially LADH, which dissociates in aqueous solution above 1500 bar and as a re-sult is inactivated under these conditions, shows a dramatic increase in activity by incorpo-rating the enzyme in micelles. Additionally, the formation of rod-like micelles under pressure benefits the optimum contact between the surface of the enzyme and micelle, which influ-ences strongly the catalysis of the reaction.en
