dc.description.abstractIn der vorliegenden Masterarbeit wurden die Filamentstrukturen der Plasmodium Aktine untersucht. Hierzu wurde die Transmissionselektronenkryomikroskopie und Methoden der Einzelpartikelanalyse verwendet, welche seitens der Softwarepakete EMAN/EMAN2, SPARX und CTFFIND4 bereitgestellt wurden. Zur Verbesserung der Auflösung des Aktin 1 wurden verschiedene Methoden der digitalen Datenaufreinigung angewendet und die Maskierung optimiert. Außerdem wurden die helikalen Symmetrieparameter dieser Isoform bestimmt. Für die Filamentstruktur von Plasmodium falciparum Aktin 1 konnte eine Auflösung von 8-9 Å und für die Struktur von Plasmodium berghei Aktin 2 eine Auflösung von 12-14 Å erzielt werden. Anhand eines Vergleichs der Struktur des Aktin 1 mit der eines kanonischen Aktins konnte zudem die Hypothese aufgestellt werden, dass strukturelle Unterschiede die Ursache der besonderen Eigenschaften dieser Isoform sind. Da zur Bestätigung dieser Hypothese eine höher aufgelöste Struktur benötigt wird, befasst sich diese Masterarbeit auch mit möglichen, die Auflösung limitierenden, Faktoren und nennt Methoden mit denen die Auflösung weiter verbessert werden könnte.de
dc.description.abstractIn the presented master thesis the filament structure of Plasmodium was analyzed. For this purpose, cryo transmission electron microscopy and single particle analysis methods, which are provided by the software packages EMAN/EMAN2, SPARX and CTFFIND4, were used. In order to improve the resolution of actin 1 different methods, which can be used to clean the dataset, were made use of. Additionally, the applied mask was optimized and the helical symmetry parameters of this isoform were determined. For the filament structure of Plasmodium falciparum actin 1 and Plasmodium berghei actin 2 a resolution of 8-9 Å and 12-14 Å respectively was achieved. Furthermore, by comparing the structure of actin 1 with one of a canonical actin the hypothesis could be proposed that structural differences cause the extraordinary properties of this isoform. As a high-resolution structure is needed to confirm this hypothesis, this master thesis also deals with factors that possibly can limit the resolution and suggests methods which could be used to further improve the resolution.de
