Abstract
In dieser Arbeit werden Methoden zur diskontinuierlichen Bestimmung von Methanol und Ethanol sowie zur kontinuierlichen Bestimmung von Ethanol in Außenluft beschrieben. In einer sogenannten Glas-Wendel wird die Außenluft in einer wässrigen Lösung angereichert und anschließend enzymatisch bestimmt. Methanol und Ethanol werden mit Hilfe des Enzyms Alkoholoxidase oxidiert, das Reaktionsproduktes H2O2 wird fluorimetrisch detektiert. Ethanol konnte zusätzlich mit Hilfe des Enzyms Alkoholdehydrogenase und des Coenzyms Nicotinamid-Adenin-Dinucleotid (NAD+) bestimmt werden, da dieses als Absorberlösung in der Glas-Wendel fungieren kann. Mit den beschriebenen Analysensystemen konnten für die Alkoholoxidase-Reaktion Nachweisgrenzen für Methanol von 6,03 ppbv und für Ethanol von 21,10 ppbv erreicht werden, für die Alkoholdehydrogenase-Reaktion betrug die erzielte Nachweisgrenze für Ethanol 2,38 ppbv. Die entwickelten Probenahmesysteme wurden während einer Meßkampagne im April/Mai 1996 in São Paulo und Cubatão/ Brasilien zur Bestimmung von Methanol und Ethanol in Außenluft eingesetzt.
In this work a method is described for the discontinuous determination of methanol and ethanol and for the continuous determination of ethanol in ambient air. A scrubber-coil is used for sampling and enrichment of the analyte from air in an aqueous solution, which is analysed enzymatically. The measurement of methanol and ethanol is based on the use of the enzym alkoholoxidase, the product H2O2 can be detected by fluorimetry. It is further possible to determine ethanol by the enzym alcoholdehydrogenase and the coenzym nicotinamide adenine dinucleotide (NAD+), which can be used as a reagent solution in the scrubber-coil. The detection limit of the systems were found to be 6,03 ppbv for methanol and 21,10 ppbv for ethanol with the enzym alcoholoxidase and 2,38 ppbv for ethanol by using the enzym alcoholdehydrogenase. The applicability of the developed methods were demonstrated during a field campain in São Paulo and Cubatão/Brazil in april/may 1996.
