Abstract: Im Jahr 2004 wurde das Polyketid (‒)-Lytophilippin A, welches eine eindrucksvolle Struktur und große Anzahl von stereogenen Kohlenstoffatomen (17) aufweist, durch Řezanka, Hanuš und Dembitsky aus dem Nesseltier Macrorhynchia philippina isoliert und charakterisiert. 2011 veröffentlichten Lee et al. die erste Totalsynthese von (‒)-Lytophilippin A und stellten dabei über den Vergleich der NMR-Daten fest, dass die von Řezanka et al. postulierte Struktur oder Konfiguration fehlerhaft sei. Ein Hinweis von Hodgson und arbeitskreisinterne Untersuchungen der Daten von Řezanka führten zu der Überlegung, dass der Fehler der Konfigurationszuordnung wahrscheinlich im Tetrahydrofuranring-Bereich (C11 bis C15) angesiedelt sei. Aus diesem Grund wurde (11R,13S,14R,15R)-Lytophilippin A als eines der möglichen Diastereomere identifiziert und über eine Synthesestrategie, welche potenziell alle möglichen Diastereomere des Tetrahydrofuranring-Bereichs des Naturstoffs zugänglich machen kann, hergestellt. Der Vergleich der NMR-Daten von (11R,13S,14R,15R)-Lytophilippin A mit den Daten des von Řezanka et al. isolierten (‒)-Lytophilippin A zeigte umfangreiche globale und lösungsmittelunabhängige Abweichungen der 1H- und 13C-NMR chemischen Verschiebungen, was auf eine tiefsitzende und globale Fehlzuordnung der Konfiguration des Naturstoffs hinweist.
In 2004, the polyketide (‒)-lytophilippine A, which has an impressive structure and a large number of ste-reogenic carbon atoms (17), was isolated and characterized by Řezanka, Hanuš und Dembitsky from the cnidarian Macrorhynchia philippina. In 2011, Lee et al. reported the first total synthesis of (‒)-lytophilippine A (I) and noticed that the structure or configuration postulated by Řezanka et al. was erroneous by comparing the NMR data. A hint from Hodgson and in-house investigations of the data from Řezanka led to the consideration that the error of the configuration assignment is probably located in the tetrahydrofuran ring region (C11 to C15). For this reason, (11R,13S,14R,15R)-lytophilippine A has been identified as one of the possible diastereomers and was produced through a synthetic strategy that can potentially access all possible diastereomers of the tetrahydrofuran ring region of the natural product. The comparison of the NMR data of (11R,13S,14R,15R)-lytophilippine A with the data of the (‒)-lytophilippine A isolated by Řezanka et al. showed extensive global and solvent independent deviations of the 1H and 13C NMR chemical shifts, indicating a deep-seated and global misallocation of the natural product configuration.
